摘    要:
通过邻苯三酚自氧化法测定酶活,从渤海海域中筛选出一株高产超氧化物歧化酶(SOD)酵母菌,其SOD酶活达4 185.67 U/g湿菌体。通过形态学、生理生化鉴定,并结合26S rDNA系统发育分析,初步确定该菌株属于海洋胶红酵母(Rhodotorula sp. CD-008)。粗酶液经SDS-PAGE、Native-PAGE、酶活性染色分析,得到酶的分子质量为37.5 ku,推测SOD类型为Cu/Zn-SOD。为研究超氧化物歧化酶在生物体内抗氧化作用以及在医、药、食品等方面的应用奠定了基础。

Abstract:
A high superoxide dismutase (SOD) yield strain was isolated from the Bohai Sea by measuring the enzyme activity in pyrogallol autoxidation, with enzyme activity of 4 185.67 U/g wet cells. After morphological, physiological, biochemical identification and 26S rDNA phylogenetic analysis, the strain was identified as Rhodotorula sp. CD-008. The crude enzyme liquid was analyzed by SDS-PAGE, Native-PAGE, enzyme activity staining, and the SOD was presumed to be Cu/Zn-SOD with molecular mass of 37.5 ku. The experiment made a foundation for the antioxidant effect of SOD in vivo and the application in clinic, medicine, food and other aspects.
