Abstract:In this study,a method for determination of dihydromyricetin,efficiency ingredient of health foodsAmpelopsis grossedentata,was established by UPLC-UV.The optimized extraction conditons were listed: the health foods produced from the plant were extracted with ethanol(65%) under the heat reflux extraction,while the health foods dispensed from the crude extracts were extracted with ethanol(65%) under the ultrasound extraction.The optimized chromatographic conditions were as follows: the seperation column was Acquity UPLCTM BEH C18 100 mm×2.1 mm(i.d),1.7μm;the mobile phase was acetrile-2% acetic acid solution(15+85 V+V) with the flow rate of 0.3 mL/min;the contents of acetrile in the mobile phase were 15% hold for the first 2 min,increased linearly to 20% within 1 min,then increased linearly to 90% within 4 min and hold for 1 min,finally decrease linearly to 15% within 3 min and hold for 6 min;the detector waverlength was 294nm;the column temperature was25℃.The assessment results of the analytical method were showed: the standard solution of dihydromyricetin showed good linear in 2.0 μg/mL~200.0 μg/mL,the average recoveries were 89.6%~96.1%,the RSD were 4.1%~8.0%(n=5),the LOD was 2.0ng.the analytical method showed satisfsctory results when applied in real samples.
