低交联两性树脂吸附蛋白质与盐浓度关系的研究

摘    要:研究低交联度两性树脂吸附刀豆蛋白与盐浓度的关系。通过盐浓度对吸附的动力学和热力学影响的研究,结果表明,在中性磷酸盐缓冲中,两怀树脂吸附刀豆蛋白,有盐(0.4mol/L NaCl)的比无盐的扩散系数提高将近一半,且量增加其吸附等温线的效率在最初始部分高于无NaCl时的;随着盐浓度(0 ̄1mol/L NaCl)的提高,树脂对蛋白的吸附最也变大,盐浓度提高时树脂听增大与蛋白吸附量的增加呈线性关系,内盐型

Abstract:The relation between salt conc. and adsorption of jack bean proteins onto the amphoteric resin was studied in this paper. The amphoteric resin was in innersalt form. The results of adsorption experiments show that when the proteins are adsorpted into the amphoteric resin in the neutral phosphate buffer solution, the diffusion coefficient of protein is near twofold in 0.4 mol/L NaCl phosphate buffer solution than that in solution without salt, and the adsorption capacity of protein also rises at the same time, its slope of adsorption isotherm in the salt solution is more than that in the solution without NaCl. When the salt conc.(at 0~1 mol/L NaCl) is increased, the adsorption capacity of resin to proteins will rise, and the adsorption capacity of protein has linear relatoin with the swelling volume of resin. The adsorption mechanism is the molecular sorption by breakdown of innersalt bond, it is the same sorption mechanism of small molecular high valent cation.
