SPECTROPHOTOMETRIC DETERMINATION OF NITROGEN IN WORT |

Abstract: | A simple spectrophotometric method is described for the determination of nitrogen in unhopped wort and end-fermented wort. After a 100-fold dilution, the extinctions at 215 mμ, and 225 mμ, are measured. The nitrogen content (mg. per litre) is calculated from the linear regression equations, Nwort = (2321 × Δ E) — 16 and Nend-fermented wort = (2474 × ΔE) — 161. Because of its simplicity and rapidity the method has considerable advantages over the classic Kjeldahl method. The extinctions at these low wavelengths are less dependent on the nature of the protein and are less disturbed by non-protein compounds than is the extinction at the more usual wavelength of 280 mμ. The formulae cannot, however, be applied to hopped wort and beer because of interference by hop constituents. |
