细菌海洋酶提取虹鳟鱼皮胶原蛋白的工艺研究

摘    要:胶原蛋白是一种具有丰富营养价值和生物功能的结构蛋白质,广泛应用于食品、药品行业。本研究以单因素试验为基础,以虹鳟鱼皮胶原蛋白提取率为指标,采用正交试验对细菌海洋酶用量、酶解温度、酶解pH、酶解时间进行了优化,探讨了酶法提取虹鳟鱼皮胶原蛋白的新工艺。结果表明:细菌海洋酶M的提取率优于细菌海洋酶L;当酶剂量为1.2%、酶解时间80 min、酶解温度55℃、酶解pH 9时,虹鳟鱼皮胶原蛋白提取率最佳,为26.1%。以废弃虹鳟鱼皮作试验材料,可减少环境污染,节约资源,并提高水产品废弃物的附加值。

Abstract:Collagen was one kind of structural proteins with abundant nutrition and biological function,which was widely used in food and medicine.The dosage of bacteria marine enzyme,enzymolysis temperature,enzymolysis pH and enzymolysis time were optimized by orthogonal test,based on single factor test and by using the extraction rate of collagen from rainbow trout skin as index.The results indicated that the extraction rate by bacteria marine enzyme M was superior to the one of L,and the extraction rate 26.1% was the highest under the condition of dosage of enzyme 1.2% when the enzymolysis duration 80 min,the temperature 55 ℃,and the pH 9.It could reduce environmental pollution,conserve resources,and improve the added value of fish waste by using the discarded rainbow trout skin as test materials.
