酶膜耦合制备高活性ACE抑制玉米肽及结构研究

摘    要:为了充分提高酶解效率及获得高活性的ACE抑制玉米肽,采用酶解与膜分离耦合技术连续化制备高活性ACE抑制玉米肽,考察了该制备过程中产品的质量和稳定性,比较了不同分子质量级分玉米肽的ACE抑制活性,以及采用HPLC-MS/MS对几种玉米肽的结构进行了解析。结果显示:在连续制备5 h内,所得产品的ACE抑制活性及肽含量较稳定,ACE抑制率在84%左右(最高达到89.82%),其中Mw<3 ku级分玉米肽其ACE抑制活性最高(IC50=0.29 mg/mL);解析出了3种玉米肽的结构,其氨基酸序列分别为QQLLPF、QQFLPF和QFLPF。试验表明:连续化酶膜耦合技术制备的ACE抑制玉米肽产品质量高且稳定,其中Mw<3 ku的玉米肽级分的ACE抑制活性最高;解析出的3种玉米肽均具有ACE抑制肽的结构特点。
