α-环乙基乙酰乙酰苯胺类化合物的合成研究

摘    要:β-二羰基化合物具有自己独特的反应,而含有活泼亚甲基的乙酰乙酰苯胺类化合物合成的小环化合物在有机合成中有广泛的应用,如用于合成药物中间体或药物成环试剂等。以二烯酮为原料合成乙酰乙酰苯胺,再进一步与二溴代烃反应制得α-环乙基乙酰乙酰苯胺,为环类化合物及其合成拓宽思路。探讨影响α-环乙基乙酰乙酰苯胺收率的因素,实验表明,当反应温度20℃,反应时间3h,反应底物:碱:二溴代烃=1:2.2:1,反应溶剂为DMF时反应收率最佳,可达78%。

Abstract:The β-dicarbonyl compound has its own unique reaction, and the small ring compounds which are synthesized by acetyl aeetanilide compounds, which contain active methylene, have wide applications in organic synthesis, such as intermediates for synthetic drugs and cyclization reagent, etc. Acetyl acetanilide was synthesized using ketene as raw material, and then reacted with dibromide to synthesize or-link ethyl acetyl monoacetylaniline. The factors which affected the yield were studied, when the reaction temperature was 20℃, time was 3h, reagent : alkali : dihromide were 1:2.2:1 and using DMF as solvent, the yield could reach 78%.
