荷叶中荷叶碱的高效液相色谱检测方法及其提取工艺研究

摘    要:通过正交试验探讨用乙醇回流提取荷叶中荷叶碱的最佳工艺,并建立荷叶碱含量测定的高效液相色谱测定方法。结果表明,用75%的乙醇在90℃温度下按固液比为1∶20(m∶V)回流提取3.5h,荷叶碱回收率高于95%;高效液色谱检测其含量在43.16~258.96μg/mL有很好的线性关系,相关系数达0.999 3,最低检出量为0.103 4μg/mL。说明用该方法提取荷叶碱可行,测定结果稳定、精确。

Abstract:The best technique of extracting nuciferine from lotus leaves by ethanol was determined by orthogonal experiment L(4 4).The method of testing nuciferine concentration in lotus leaves by HPLC was developed.The lotus leaves were extracted by using the solid/liquid rate of 1/20(m∶V) and alcohol of 75% at 90℃for 3.5h.The linearity relationship of nuciferine obtained from HPLC fitted very well in the range from 43.16 μ g/mL to 258.96 μ g/mL of nuciferine when it was dissolved in methanol.The relationship coefficient was 0.999 3.The detection limit was 0.103 4 μ g/mL.The extracting method was feasible technique of nuciferine from lotus leaves.The HPLC method test results were stable and precise.
