大孔吸附树脂纯化苦丁茶总皂苷的研究

摘    要:优化大孔吸附树脂纯化苦丁茶总皂苷的工艺参数。分别用AB-8、NKA-9、S-8、X-5、D101、HP-20对其进行静态吸附与解吸试验,筛选出效果较好的D101树脂。通过对D101树脂分离苦丁茶总皂苷的动态试验。结果表明,优化的吸附条件:流速为2BV/h时,料液的pH值和浓度分别为6和14.47mg/mL;优化的洗脱条件:流速为2.5BV/h,先用2BV水洗脱去杂,然后用3BV 70%乙醇水溶液进行洗脱并收集洗脱液、浓缩、冷冻干燥,得到总皂苷含量为68.9%粉末,纯化倍数为1.77。D101型大孔树脂纯化苦丁茶总皂苷的方法可行,具有良好的应用前景。

Abstract:To optimize the technological parameters of purifying total saponin from Ilex kudingcha c.J.Tseng by macroporous resins.the static adsorption and desorption experiments were made by AB-8,NKA-9,S-8,X-5,D101,HP-20,respectively.D101 resin is chosen for its good effect.Dynamic experiment about purification of total saponin was investigated by D101 resin.the results showed that optimal adsorption conditions were the concentration of the raw material with six of pH value was 14.47mg/mL when flow velocity was 2BV/h and optimal elution conditions were elution velocity was 2.5BV/h,firstly eluted with 2BV of water to remove impurities and then with 3BV 70% alcohol with aqueous solution to get eluent,which was collected and concentrated and freeze-dried to get powder with total saponins content 68.9% and purification times 1.77.Purification of total saponin from Ilex kudingcha c.J.Tseng by macroporous resins D101 is feasible and has good application prospect.
