司帕沙星-钇-十二烷基硫酸钠荧光体系的研究及应用

摘    要:用荧光光谱法研究了钇(Y3+)和十二烷基硫酸钠(SDS)与司帕沙星的相互作用,发现司帕沙星能同Y3+形成稳定的络合物,SDS对络合物有较强的增敏作用,据此建立了用稀土Y3+作为荧光探针检测微量司帕沙星的新方法。结果表明;在pH 8.2的醋酸铵-氨水缓冲溶液中,Y3+浓度为1.0×10-5mol·L^-1、SDS浓度为3.0×10-4mol·L^-1时,司帕沙星的浓度在0.2×10-6~6.0×10-6mol·L^-1和0.6×10-5~2.5×10-5mol·L^-1范围内与体系荧光增敏强度呈良好的线性关系,相关系数分别为0.995 2和0.995 8,检出限为4.16×10-8mol·L^-1。应用于司帕沙星片剂中司帕沙星含量的测定,结果令人满意。

Abstract:Interaction between yttrium(Y3+)、sodium dodecyl sulfate(SDS) and sparfloxacin was investigated by fluorescence spectroscopy.The result shows that a stable complex is formed by sparfloxacin and Y3+,SDS has a sensitization to the complex.A novel method for determining trace sparfloxacin with Y3+ as fluorescent probe is established.The optimum reaction conditions are that Y3+ concentration is 1.0×10-5mol·L^-1,SDS concentration is 3.0×10-4 mol·L^-1 in ammonium acetate-aqueous ammonia buffer solution at pH 8.2.The quenching fluorescence intensity is linearly proportional to the concentration of sparfloxacin from 0.2×10-6 to 6.0×10-6 mol·L^-1 and 0.6×10-5 to 2.5×10-5 mol·L^-1,correlation coefficient R=0.995 2 and 0.995 8.The detection limit for sparfloxacin is 4.16×10-8 mol·L^-1.The method is applied to determination of sparfloxacin contents in pill with satisfactory results.
