储存喷气燃料中悬浮物与真菌的相关性研究

摘    要:采用显微镜观测、能量色散X射线分析(EDX)、颗粒数测定、真菌培养及鉴定和聚合酶链式反应(PCR)扩增及凝胶电泳检测手段,研究了储存喷气燃料中悬浮物与真菌的相关性。结果表明,在光学显微镜下能看到储存喷气燃料中的悬浮物有如真菌菌丝特有的横隔结构;悬浮物的元素主要为C、N、O,说明其主要由有机质组成。含有悬浮物的储存喷气燃料用0.45 μm滤膜过滤(除去孢子)并加入去离子水,放置75 d后各级颗粒数无明显变化,而未过滤(含有孢子)者放置75 d后各级颗粒数显著增加。悬浮物样品在沙保氏、PDA培养基上能长出真菌菌落,其菌丝的尺寸、结构与悬浮物的相一致。悬浮物样品经PCR扩增并用琼脂糖凝胶电泳检测,均得到与真菌DNA提取液相同的条带,表明悬浮物中含有真菌DNA。因此,真菌与喷气燃料中悬浮物的形成密切相关。

Study on the Relativity Between Fungus and Suspended Matters in Stored Jet Fuel

Abstract:The microscope, energy dispersive X-ray spectroscope(EDX), particle determination, fungal culture and identification, polymerase chain reaction (PCR) and agarose electrophoresis analysis were used to research the relationship between fungus and suspended matters in stored jet fuel. The results showed that the septa were found in suspended matters of stored jet fuel, which is peculiarly in fungus hyphae. The elements of suspended matters were mainly C, N, O, meaning that the suspended matters were composed of organic matter. After adding deionized water, the particle number of the fuel without filtrating(having conidia) increased significantly, while the one after filtrated by 0.45 μm millipore filter(getting rid of conidia) didn’t change evidently in 75 d. Fungal culture of suspended matters was carried on in Sabouraud medium and PDA medium to get the fungus colony. It is found that the fungus hyphae had the same size and structure as the suspended matters. PCR amplification of suspended matters got the same bands as the fungus DNA under agarose electrophoresis analysis, showing that fungus DNA was contained in both of them. So, the fungus is proved to be closely related to suspended matters in stored jet fuel.
