亚临界水中大豆渣水解制备氨基酸工艺研究

摘    要:豆渣是大豆加工过程中的主要副产物。今在亚临界水中对豆渣水解制备氨基酸的生产工艺进行了研究,为大豆的综合利用开辟了一条新的途径。利用氨基酸分析仪对水解产物中的氨基酸进行定性和定量分析,实验结果发现,大豆渣水解后可得到17种氨基酸。考察了反应温度、反应时间、物料浓度以及CO2分压对氨基酸总收率的影响,并进行正交试验。研究结果表明,亚临界水解法高效快速,工艺简单,CO2气体的加入有利于增加氨基酸总收率,较佳的水解工艺条件为:T=200℃,P(CO2)=3.0 MPa,w=5.0 mg mL 1,t=15 min,氨基酸的总收率为14.25%。

Abstract:Hydrolysis of soybean residue,one of major by-products of soybean processing,in sub-critical water to produce amino acids was conducted in this paper.The quality and quantity analysis of amino acids in hydrolysate were carried out by Amino Acid Analyzer(Biological Liquid Chromatography),and 17 kinds of amino acids were obtained.The effects of reaction temperature,time,row material concentration and pressure of carbon dioxide on the yield of amino acids were investigated.Based on the experimental results,the technological optimum of the hydrolysis process was studied by using L9(34) orthogonal test method.The results show that the sub-critical water hydrolysis process has the advantages of high efficiency,simple process and friendly to environment.It was found that the optimum hydrolysis conditions for amino acids preparation from soybean residue in sub-critical water are as follows: reaction temperature 200℃,pressure(carbon dioxide) 3.0 MPa,row material concentration 5.0 mg mL 1 and reaction time 15 min.Under these conditions,the yield of amino acids is 14.25%.
