摘    要:利用X射线微区分析,对固定化青霉素G酰化酶的活进行了定位分析,青霉素G钠作为底物,FeCl3作为捕捉剂,底物经固定化青霉素G酰化酶水解产生苯乙酸,后者与捕捉剂反应生成沉淀,可以确定化青霉素G酰化酶的催化活性部位,本工作对不同酶活的固定化青霉素G酰化酶进行了形态分析与活性定位的研究,发现酶活的部位集中分布在载体的表面上,且分布不均匀,该方法可靠,准确。

Abstract:Localization of activity of immobilized penicillin G acylase was studied by X ray microanalysis. Benzylpenicillin sodium and ferric chloride severed as substrate and capture respectively. Substrate was hydrolyzed by immobilized penicillin acylase to produce phenylacetic acid precipitating with capture. Precipitation deposited active site of immobilized penicillin G acylase. The morphological analysis and localization of activity of immobilized penicillin G with different activities were studied. It was found that the most of active enzyme distributed irregularly on the surface of carrier. The method was practicable and reliable.
