摘    要:采用液相色谱内标法建立了环十五内酯合成中的重要化合物1,15-十五碳二元酸的定量分析方法。通过实验确定了最优色谱条件:色谱柱C18(4.6×250mm),以月桂酸为内标物,流动相为甲醇∶乙腈∶水=80∶5∶15(0.1%三氟乙酸),流速为1.2mL/min,检测波长为210nm。方法的相关性好(γ〉0.9990),样品测定的相对标准偏差RSD〈1%,加标回收率为99.25%,检出限为4.543×10-4g/(s·L)。

Abstract:A method of reversed phase high performance liquid chromatography (HPLC) with internal standard was applied to the quantitative analysis of 1,15-pentadecanedioic acid, which was an important material in the synthesis of cyclopentadecanolide. Optimum conditions were determined as follows: column packed with C18(4.6×250 mm), lauric acid as internal standard.The mobile phase consists of a mixture of methanol, acetonitrile and water(80∶5∶15, 0.1%trifluoroacetic acid) with a flow rate of 1.2 mL/min and UV detection performed at 210nm. Desired relativity(r0.9990)and analytic precision (RSD 1% ) were obtained by the method with average recovery rate of 99.25% and sensitivity of 4.543×10-4 g/(s·L).
