1-杂氮吩噻嗪的合成研究

摘    要:以2-氯吡啶和苯胺为原料,经缩合、硫粉环合制得1-杂氮吩噻嗪。较佳合成工艺条件为:n(2-氯吡啶)∶n(苯胺)=1∶1,反应温度180℃,2-苯胺基吡啶收率95.6%,纯度99.5%(HPLC);二苯醚为溶剂,碘为催化剂,n(2-苯胺基吡啶)∶n(硫粉)=1∶1.8,m(2-苯胺基吡啶)∶m(二苯醚)∶m(碘)=1∶1∶0.05,反应温度270℃,1-杂氮吩噻嗪收率71.3%,纯度99.2%(HPLC);总收率68.2%。产物结构经IR、1 H NMR确证。

Abstract:1-Azaphenothiazine was an important antihistamine and antipsychotic intermediate.It was prepared from 2-chloropyridine and aniline as starting material via two steps including condensation and cyclization with sulfur.The process conditions were optimized: n(2-chloropyridine)∶n(aniline)=1∶1,reaction temperature was 180 ℃,2-phenylaminopyridine was obtained with yield of 95.6% and purity 99.5%(HPLC);Diphenylether as solvent,I2 as catalyst,n(2-phenylaminopyridine)∶n(sulfur)=1∶1.8,m(2-phenylaminopyridine)∶m(diphenyl-ether)∶m(I2)=1∶1∶0.05,reaction temperature was 270 ℃,1-azaphenothiazine was prepared with yield of 71.3% and purity 99.2%(HPLC);The total yield was 68.2%.The product was confirmed by IR and 1H NMR.
