水通道蛋白8在人胎盘和胎膜中的表达

目的:探讨水通道蛋白8(aquaporin 8,AQP8)在正常人胎膜及胎盘中的表达情况。方法:采集6例正常足月剖宫产的胎膜和胎盘组织,采用逆转录聚合酶链反应(RT-PCR)技术检测AQP8mRNA在胎膜、胎盘中的表达,用免疫组织化学的方法检测AQP8蛋白在胎膜、胎盘中的定位表达。结果:AQP8mRNA在羊膜、绒毛膜和胎盘均有表达,AQP8蛋白主要表达于羊膜上皮细胞、绒毛膜滋养细胞和胎盘合体滋养细胞的胞浆和胞膜中。结论:AQP8在羊水平衡的调节和母儿液体交换过程中可能起重要作用。

Expression of aquaporin 8 in human placenta and fetal membranes

Objective:To detect Aquaporin 8(AQP8) expression in normal human fetal.membranes and placenta.Methods:Fetal membranes and placenta were collected from 6 women with intact membranes who underwent elective cesarean sections at term.RT-PCR was used to detect expression of AQP8mRNA,immunohistochemistry was used to detect expression of AQP8 protein.Results:AQP8 mRNA was detected in amniotic,chorion and placenta.Immunohistochemical results:showed AQP8 protein was mainly localized in the cytomembrane and cytoplasm of epithelial cell of amnion,trophocyte of chorion and syncytiotrophoblast of placenta.Conclusions:AQP8 may play an important role in amniontic fluid balance as well as maternal-fetal fluid balance.
