梅花鹿茸多糖提纯

摘    要:以梅花鹿鹿茸为原料,经提取、分离、纯化得到鹿茸多糖(PAPS)粗样品,将粗品在DEAE-Cellulose 32和Sephadex G-75柱上进一步纯化,得到PAPS样品.经SDS-聚丙烯酰胺凝胶电泳检测,PAPS的相对分子量为70 kDa-80 kDa,采用苯酚-硫酸法测得总糖含量为825.51μg/mg,Folin-酚法测得蛋白质含量为20.27μg/mg,硫酸钡比浊法测得硫酸基含量为58μg/mg.红外光谱分析表明:PAPS具有明显的多糖特征吸收峰.

Abstract:Pilose antler polysaccharides(PAPS) were obtained from pilose antler by the method of extraction,seperation and purification.The sample of PAPS was purified with the column chromatography on DEAE-Cellulose 32 and Sephadex G-75.Its molecular weight is about 70~80 kDa detected by SDS-polyacrylamide gel electrophoresis,the content of total sugar is 825.51 μg/mg determined by phenol-sulfuric acid method,the content of protein is 20.27 μg/mg determined by Foiln-hydroxybenzene method,and the content of sulfate radical is 58 μg/mg determined by barium sulfate turbidity.The sample of PAPS exhibits typical absorption of polysaccharide characterized by IR analysis.
