二次正交旋转组合设计优化青霉产α-葡萄糖苷酶培养基

摘    要:优化青霉产α-葡萄糖苷酶培养基的3个因素配比.应用DPS v7.05统计分析软件中的三元二次正交旋转组合设计方案进行试验设计.通过二次多项回归方程寻优,当麦芽糖5.5%、蛋白胨1.25%、麸皮汁:土豆汁3∶2,此时的酶活力达到最大4.23u/ml,比优化前酶活力2.49u/ml提高了41.1%.通过产酶验证试验得出实验值与模型预测值基本符合,说明二次正交旋转组合设计可用于产酶培养基组分的优化和分析.

Abstract:Optimized the formula of three ingredients of medium for Penicillium production of α-glucosidase, methods design the test by using the quadratic orthogonal rotation combination design in DPS software. Results the optimization of quadratic regression equation was in maltose 5.5%, peptone 1.25%, bran juice∶potato juice 3∶2, enzyme activity reach maximum at this time, increased 41.1% the enzyme activity than before. Conclusion quadratic orthogonal rotation combination design was suitable for the optimization of ingredients of fermentation medium.
