产蛋白酶海洋微生物的筛选鉴定及发酵

摘    要:从秦皇岛附近海域水样中筛选出了1株蛋白酶生产菌,并对其发酵条件进行了初步研究.经16S rDNA鉴定此菌株属假交替单胞菌属(Pseudoalteromonas sp.),所产蛋白酶为中性蛋白酶.液体培养基为人工海水,蛋白胨10.0g/L,酵母浸粉5.0g/L,葡萄糖5.0g/L,硝酸铵1.0g/L,无水氯化钙1.0g/L,在33℃,初始pH值6.0,50mL装液量,200r/min条件下发酵,发酵液酶活可达1140 U/mL.

Abstract:A strain of protease-producing marine microorganism was isolated from the sea-water in Qinhuangdao. The marine microorganism was identified belonging to the genus of Pseudoalteromonas sp., and the protease which produced by the strain was neutral. The optimum broth was consisted of peptone 10.0g/L, yeast extract powder 5.0g/L, glucose 5.0g/L, NH4NO3 1.0g/L, CaCl2 1.0g/L. Enzyme activity could reach 1140U/ml under the fermentation condition: fermentation temperature 33℃, initial pH 6.0, filling volume 50 ml, rotational speed 200 r/min.
