Abstract:Objective To establish a rapid method for the determination of 4-hydroxyderricin and xanthoangelol in Angelica keiskei from different places of origin and parts.Methods An HPLC-PDA was used to determine the contents of 4-hydroxyderricin and xanthoangelol with a Thermo NO.30005-254630 silica gel chromatographic column(4.6 mm×250 mm,5μm).The gradient elution was performed with 0.1%formic acid solution(a)-methanol(b),the flow rate was 1.0 ml/min,the column temperature was 40℃,the detection wavelength was 370 nm,and the injection volume was 5μl.Results The results showed that the linear range of 4-hydroxyderricin was 4.95-495μg/ml with the correlation coefficient R2 of 0.9993.The average recovery was 98.4%and the minimum detection limit was 2μg/g.The linear range of xanthoangelol was 5.65-565μg/ml with the correlation coefficient R2 of 0.99988.The average recovery was 102.3%and the minimum detection limit was 2μg/g.The effective components of 4-hydroxyderricin and xanthoangelol in 18 samples of Angelica keiskei from 6 areas were detected.Conclusion This method is simple in sample pretreatment and suitable for the rapid detection of functional components of Angelica keiskei.From different regions,the contents of 4-hydroxyderricin and xanthoangelol in Angelica keiskei in southern China ware significantly higher,but the contents of samples in Jiangsu province ware significantly lower.4-Hydroxyderricin and xanthoangelol ware mainly distributed in stems and roots,with the highest level in roots and the lowest level in leaves.This study provides a reference for the industrialization of the extraction of functional components from Angelica keiskei.
