阿维菌素产生菌培养条件的优化

摘    要:对阿维链霉菌(Streptomyces Avermitilis)3~6的培养条件进行优化。结果表明:斜面培养基成分中酵母粉的产地、斜面种子的培养周期及棉塞的重量对斜面种子的质量及发酵结果的影响程度较大,经分析,得到最适酵母粉为湖北产,最佳培养周期为8d,最适棉塞重量为7g。通过正交实验优化了发酵培养基配方,发酵产量可提高7.35%。此外,还研究了前体物质丙酸钠的添加对发酵结果的影响,结果表明发酵过程中在培养基内加入一定浓度的前体物质丙酸钠,可以显著提高发酵产量,其中发酵24h时在培养基中加入2.5mmol/L的前体物质丙酸钠,发酵产量可提高9.89%。

Abstract:In this paper, the culture conditions of Streptomyces Avermitilis strain 3-6 were optimized. The result showed: The place of yeast extract production in slant medium, the cultural period of slant seed and the weight of tampon could influence the quantity of the slant seed and fermentation. Through analysis, it has been determined that the most suitable yeast extract production place is Hubei, the best cultural period of slant seed is 8 days, the most suitable weight of tampon is 7 g. The fermentation medium was optimized through orthogonal design, the yield was increased by 7.35%.In addition, the effect of the addition of sodium proplonate precursor on fermentation was studied. The result showed that adding the certain density of sodium proplonate in fermentation medium could obviously increase the yield of fermentation. Adding 2.5 mmol/L sodium proplonate into the medium at 24 h, the yield of fermentation was increased by 9.89%.
