大豆异黄酮精制工艺研究

摘    要:试验比较10种不同型号大孔树脂对大豆异黄酮吸附性质和不同溶剂萃取大豆异黄酮效果,确定HPD-600树脂吸附纯化大豆异黄酮最佳工艺条件如下:上样液浓度0.15mg/mL、上样液pH值4~5、上样量4.5BV、吸附流速1.0ml/min、静态吸附250min,用80%乙醇作为解吸剂,解吸流速为0.5ml/min,3.0BV解吸剂即可解吸完全。得到大豆异黄酮粗品含量为20.11%,比粗提物纯度提高7.18倍;同时得出丙酮沸点回流萃取可得到含量为42.91%大豆异黄酮产品,纯度比含量为20.11%原料提高2.13倍;乙酸乙酯和丙酮组合沸点回流萃取,可得到含量为70.36%大豆异黄酮产品,纯度比含量为42.91%原料提高1.64倍。

Abstract:Study on the absorption from Isoflavone by resin and static absorb research by ten kinds of resins and compares the effect on extraction of different solvent in the experiment,The technology of purifying isoflavone by macroporous resin HPD-600 is established.the parameters are as follows:the concentration of crude isoflavone solution is 0.15 mg/mL;its pH value is 4~5;its volume is 4.5 times as resin bed volume;crude isoflavone solution is adsorbed at the speed of 1.0 ml per minute,then adsorbed in static state for 250 minutes,desorbed with 80% aqueous ethanol at the speed of 0.5 ml per minute,its volume is 3.0 times as resin bed volume;After condensed and dried,desorption solution becomes purified product.The soybean isoflavone content of which is 20.11%,enhanced 7.18 times than raw material.the return extraction of acetone boiling point that has been allowed to obtain the content is 42.91% soybean isoflavone product,the purity enhances 2.13 times compared to the content 20.11% raw materials;the return extraction of ethylacetate and acetone combination boiling point,may obtain the content is 70.36% soybean isoflavones product,the purity enhances 1.64 times compared to the content 42.91% raw materials.
