东方栓孔菌三萜的大孔树脂纯化工艺优化及对小鼠酒精性肝损伤的保护作用

摘    要:为得到纯度较高的东方栓孔菌三萜化合物,采用大孔吸附树脂法对东方栓孔菌三萜粗提物进行纯化。以急性酒精性肝损伤小鼠为模型,评价了东方栓孔菌三萜的保肝作用。结果表明,X-5树脂对东方栓孔菌三萜具有较好的静态吸附与解吸性能,三萜在树脂表面的吸附符合Langmuir模型。东方栓孔菌三萜在X-5树脂上的较优纯化工艺为:上样质量浓度为2.34 g/L,上样流速为2 BV/h;洗脱时选用70%乙醇,洗脱流速为2 BV/h。东方栓孔菌三萜经X-5树脂纯化后,纯度为83.30%,是纯化前的3倍,表明X-5树脂纯化东方栓孔菌三萜效果较佳。东方栓孔菌三萜能极显著降低酒精性肝损伤小鼠的血清丙氨酸氨基转移酶和天冬氨酸氨基转移酶含量(p<0.01),极显著提高小鼠肝脏超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶活性,降低丙二醛含量(p<0.01),表明东方栓孔菌三萜可缓解酒精致小鼠急性肝损伤。

Abstract:Macroporous adsorption resins were utilized for purification of triterpenes from the crude extracts of Trametes orientalis. Hepatoprotective effect of triterpenes from Trametes orientalis was evaluated by an alcohol-induced liver injury model in mice. The results revealed that X-5 resin offered the best adsorption and desorption properties for triterpenes from Trametes orientalis. The adsorption equilibrium datas of triterpenes on X-5 resin were fitted to the Langmuir model. Optimal purification parameters were as follows:Loading concentration, loading flow rate, ethanol concentration, and elution flow rate were 2.34 g/L, 2 BV/h, 70% and 2 BV/h, respectively. The purity of triterpenoids purified by X-5 resin was 83.30%, which was 3 times higher than crude extracts. X-5 resin had excellent performance and could be applied to purify triterpenes from Trametes orientalis. The increased serum alanine aminotransferase and aspartate aminotransferase levels of mice after alcohol treatment were inhibited by the triterpenes administration extremely significant (p<0.01). Furthermore, the triterpenes could effectively increase the superoxidase dismutase, catalase and glutathione peroxidase activities, and inhibit the increase in malondialdehyde level extremely significant (p<0.01). These results indicated that the triterpenes from Trametes orientalis might be of therapeutic value in ameliorating the hepatotoxicity caused by alcohol.
