摘    要:目的建立一种鲍鱼内脏多糖的双水相盐析萃取—酶解法的提取工艺,并对多糖组成进行分析。方法样品经双水相盐析萃取,木瓜蛋白酶酶解醇沉后获得粗多糖。考察鲍鱼内脏干粉的加入量和酶对多糖得率的影响。结果鲍鱼内脏多糖双水相盐析萃取的最适条件为:乙醇30%(V:V)、碳酸钠12%(m:V)和鲍鱼内脏冻干粉加入量7 g/100 mL,酶解最适用酶为木瓜蛋白酶。在此条件下,鲍鱼内脏多糖得率为3.76%。结论经液相色谱分析,该多糖由甘露糖、氨基葡萄糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、氨基半乳糖、葡萄糖、半乳糖、木糖和岩藻糖组成,其质量比为3.54:2.41:14.75:1.00:1.25:6.79:84.04:7.79:3.38:10.83。

Abstract:Objective To establish the extraction process of polysaccharide from abalone viscera by salting-out extraction coupled with enzymolysis, and to analyze the composition of polysaccharide. Methods The samples were extracted by methods of salting-out extraction, and coarse polysaccharides were obtained after papain enzymolysis and ethanol precipitation. The influence of the amount of dried powder of abalone and the effect of enzyme on the yield of polysaccharide were investigated. Results The optimal conditions for the extraction of the viscous polysaccharides of abalone were as follows: ethanol was 30% (V:V), Na2CO3 was 12% (m:V) and the amount of permafrost powder of abalone was 7 g/100 mL. The optimum proteinase for enzymolysis was papain. The polysaccharide from abalone viscera yield was 3.76% under the optimal conditions. Conclusion By using liquid chromatography, it is found that polysaccharide is composed of monosaccharides, mannose, glucosamine, rhamnose, glucuronic acid, galacturonic acid, galactosamine, glucose, galactose, xylose, and fucose, with the mass ratios of 3.54: 2.41: 14.75: 1.00: 1.25: 6.79: 84.04: 7.79: 3.38: 10.83.
