高效液相色谱法同时测定紫丁香苷、绿原酸、栀子苷和红景天苷的含量

摘    要:目的建立高效液相色谱法同时测定紫丁香苷、绿原酸、栀子苷和红景天苷的含量。方法样品经过前处理,以0.05%磷酸水:乙腈:甲醇(85:10:5,V:V:V)为流动相,等度洗脱,经反相硅胶柱(4.6 mm×150 cm,5μm)分离,并于220 nm波长检测。结果紫丁香苷在浓度1.0~51μg/m L之间呈现良好的线性关系,r=0.9995,平均回收率为99.16%~99.32%,相对标准偏差为0.10%;绿原酸在浓度5.3~27μg/m L之间呈现良好的线性关系,r=1.0000,平均回收率在99.08%~99.51%之间,相对标准偏差为0.23%;栀子苷在浓度6.1~30μg/m L之间呈现良好的线性关系,r=0.9990,平均回收率在98.16%~98.38%之间,相对标准偏差为0.13%;红景天苷在浓度5.3~26μg/m L之间呈现良好的线性关系,r=0.9990,平均回收率在98.75%~99.86%之间,相对标准偏差为0.58%。结论该方法能同时测定紫丁香苷、绿原酸、栀子苷和红景天苷含量,耗时短、操作简便、准确、重现性好,可在实验室推广。

Abstract:Objective To establish a quick and simple method for determination of Syringin(CAS:118-34-3), Chlorogenic acid(CAS:327-97-9), Gardenoside(CAS:24512-62-7), Salidroside(CAS:10338-51-9) in health foods by high performance liquid chromatography (HPLC). Methods Samples were pretreated and used positive phase silica gel or equivalent performance chromatographic column for rapid separation of Syringin, Chlorogenic acid, Gardenoside, and Salidroside with 0.05% phosphoric acid water, methanol and acetonitrile as mobile phase mixture, respectively 85%,10%,5%,and degree of elution. The detection wavelength was 220 nm. Results It is good to separated Syringin, Chlorogenic acid, Gardenoside, and Salidroside by this method. The concentration of Syringin had a good linear relationship within 0.010~0.051mg/mL (r=0.9995), the average recoveries was 98.975%~99.506%, and the RSD was 0.24%. The concentration of Chlorogenic acid had a good linear relationship within 0.0054~0.0270mg/mL (r=1.0000), the average recoveries was 99.834%~100.028%, and the RSD was 0.38%. The concentration of Gardenoside had a good linear relationship within 0.0061~0.0200mg/mL (r=0.9999), the average recoveries was 98.117%~99.338%, and the RSD was 0.50%. The concentration of Salidroside had a good linear relationship within 0.0054~0.0270mg/mL (r=0.9990), the average recoveries was 98.292%~100.024%, and the RSD was 0.59%. Conclusion The method can be used for determination of Syringin, Chlorogenic acid, Gardenoside, and Salidroside in health foods, it is quick, simple and accurate, and has a good reproducibility and clear separation, which can be popularized in the laboratory.
