Abstract:Objective To evaluate the uncertainty of determination of furosine in sterilized milk by liquid chromatography. Investigate the steps that contribute the most to the uncertainty of the final result, and control the key steps. Methods The content of furosine in sterilized milk was determined according to NY/T939-2016 "Identification of reconstituted milk in pasteurized milk and UHT sterilized milk". The mathematical model of uncertainty evaluation for determination of furosine in sterilized milk by high performance liquid chromatography was established. The sources of uncertainty in the experiment process were analyzed comprehensively, and each uncertainty was quantified and synthesized. The standard uncertainty and expanded uncertainty of synthesis of furosine in sterilized milk were calculated. Results When the content of furosine in sterilized milk was 151.0 mg/100 g protein, the expanded uncertainty of measurement results was 5.8 mg/100 g protein (k=2). Conclusion The evaluation method of medium-range uncertainty in the determination of furosine by liquid chromatography is established. Pretreatment of furosine, protein detection and measurement repeatability are the main sources of measurement errors.
