New infiltration method permits use of freeze substitution for preparation of wood tissues for transmission electron microscopy

Abstract:A new infiltration method was developed for preparation of wood tissues for transmission electron microscopy (TEM) using freeze substitution. The method included treatment of freeze-substituted samples with sodium phosphate buffer before infiltration with epoxy resin. Samples from red osier (Cornus sericea L.) and flowering dogwood (C. florida L.) were studied to assess the quality of the infiltration method. The method produced well-embedded samples with good trimming and cutting qualities. To our knowledge, this is the first report on preparation of wood tissues for TEM using freeze substitution.
