金盏菊中叶黄素的分离纯化及高效液相色谱的测定方法

摘    要:金盏菊萃取物 15g ,加入 30mL正丁醇 ,密封搅拌均匀后 (5 5℃ ) ,加入 30mLw (NaOH) =2 0 %的水溶液 ,反应 7h(70℃ ) ;皂化物中加入 30 0mL水 ,搅拌均匀 ,过滤 ,然后依次用 30mL乙醇和 5 0mL正己烷冲洗 ,得到叶黄素 ,纯度为w (C4 0 H56O2 ) =71 2 %。确定了金盏菊中叶黄素的液相色谱测定方法 ,流动相为 V(CH2 Cl2 )∶V(CH3OH)∶V(CH3CN)∶V(H2 O) =32∶38∶2 9∶1,得到了叶黄素和 15种叶黄素酯的峰 ,叶黄素的保留时间为 3 6 81min ,叶黄素酯的保留时间分布为 4 5~ 12 .0min ;在流动相中它们的最大吸收波长都为 45 4nm。叶黄素的线性范围为 0 1~ 1.0 μg ,相关系数为γ =0 9998

Abstract:Marigold extraction was homogenized with butanol under sealed condition at 55 ℃,and then was subjected to saponification with NaOH〔w(NaOH)=20%,30 mL〕 for 7 h at 70 ℃.The saponified product was mixed with 300 mL distilled water and filtered and the retained orange precipitate of lutein was washed with 30 mL ethanol and 50 mL n hexane successively.The purity of resulting lutein was w(C 40 H 56 O 2)=71 2% by HPLC analysis.An improved HPLC method was developed for the analysis of lutein in marigold,the mobile phase was V(CH 2Cl 2)∶ V (CH 3OH)∶ V (CH 3CN)∶ V (H 2O) =32∶38∶29∶1 and the detecting wavelength was 454 nm.The linear range was 0 1~1.0 μg,γ=0 9998
