Aurora-B激酶特异性抑制剂AZD1152-HQPA对人骨肉瘤细胞株U2-OS细胞的抑制作用

目的探讨激光（Aurora）-B激酶特异性抑制剂AZD1152-HQPA对人骨肉瘤细胞株U2-OS细胞的抑制作用。方法用不同浓度的AZD1152-HQPA（0、10、50、100、500 nmol.L-1）体外作用于人骨肉瘤细胞株U2-OS24、48、72 h,采用噻唑蓝（MTT）检测AZD1152-HQPA对人骨肉瘤细胞株U2-OS细胞的抑制作用;采用Western Blotting检测AZD1152-HQPA作用于人骨肉瘤细胞株U2-OS细胞前后Aurora-B激酶的表达情况。结果 AZD1152-HQPA对人骨肉瘤细胞株U2-OS细胞生长抑制作用明显,且抑制呈时间-浓度依赖性。50 nmol.L-1AZD1152-HQPA作用24、48、72 h,人骨肉瘤细胞株U2-OS细胞中Aurora-B激酶的表达随作用时间的延长而降低。结论 AZD1152-HQ-PA能抑制U2-OS细胞生长,并且抑制作用呈时间-浓度依赖性。

Inhibition Effect of AZD1152-HQPA,Specific Inhibitor of Aurora-B Kinase,on Human Osteosarcoma Cell Line U2-OS Cells

Objective To explore the Aurora-B kinase inhibition effect AZD1152-HQPA specific inhibitor on human osteosarcoma cell line U2-OS cells.Methods The human osteosarcoma cell line U2-OS cells were treated with AZD1152-HQPA at various concentration（0,10,50,100,500 nmol·L-1） in vitro for various time（24,48,72 hours）.MTT assay was used to measure the inhibition effect of AZD1152-HQPA on human osteosarcoma cell line U2-OS cells.The expression of Aurora-B kinase before and after of AZD1152-HQPA on human osteosarcoma cell line U2-OS cells were detected by Western Blotting.Results The cells growth of human osteosarcoma cell line U2-OS were inhibited effectly by AZD1152-HQPA,showing the inhibition in time-and concentration-dependent manner.The expression of Aurora-B kinase in human osteosarcoma cell line U2-OS cells,treated with AZD1152-HQPA 50 nmol·L-1 for 24,48,72 hours,was significantly reduced and the level of expression was lower as the time increased.Conclusion The cells proliferation of U2-OS were inhibited by the selective Aurora-B kinase inhibitor AZD1152-HQPA in a time-and concentration-dependent manner.
