丹参对大鼠肝组织Bcl-2表达的影响及意义

摘    要:目的探讨丹参对大鼠肝组织Bcl-2基因表达的影响和对肝纤维化的作用及其机制。方法利用CCl4诱导形成大鼠肝纤维化模型。将30只SD大鼠按随机数字表法分成纤维化模型组(A组,n=15)和丹参干预组(B组,n=15)。用RT-PCR法检测各组大鼠肝脏Bcl-2的表达,常规HE和网状纤维染色检测肝组织标本。结果大鼠肝纤维化模型成功建立。丹参干预组纤维化程度、肝细胞脂肪变性及肝组织炎症较纤维化模型组显著减轻。丹参干预组和纤维化模型组Bcl-2基因表达量分别为(0.84±0.02)%(、1.03±0.03)%。丹参干预组Bcl-2基因的表达显著低于纤维化模型组(P〈0.01)。结论丹参能够阻断CCl4诱导的肝纤维化和减少肝脂肪变性。其机制不仅直接通过抗炎作用,也可能通过调节Bcl-2基因表达从而阻断肝纤维化。

Abstract:Objective To study the effects of salvia on rat liver tissue Bcl-2 expression and liver fibrosis.Methods Hepatic fibrosis was induced in rats by subcutaneous injection of CCl4.SD rats were randomly divided into fibrosis model group(group A) and salvia intervention group(group B).The expression of Bcl-2 in liver was determined by using RT-PCR and the liver tissue samples were detected by conventional HE and reticular fiber staining.Results The rat liver fibrosis model was successfully established.Compared with group A,the liver fibrosis,inflammation and fatty degeneration in group B were significantly reduced.The expression of Bcl-2 in group B(0.84%±0.02%) was significantly lower than that in group A(1.03%±0.03%)(P〈0.01).Conclusion Salvia can effectively prevent CCl4-induced hepatic fibrosis and decrease liver fatty degeneration through the mechanisms involving its anti-inflammatory effect and the down-regulation of Bcl-2 gene.
