摘 要: | 目的观察人禽流感裂解疫苗对小鼠的免疫原性。方法将不同血凝素含量(20、30、45μg/ml)的铝佐剂和无佐剂人禽流感裂解疫苗各3批分别经腹腔注射免疫小鼠,0.5 ml/只,每批疫苗又分为1针、2针和3针组,每组10只,每针间隔1周,同时设空白对照组。首针免疫后21 d,分别采血,分离血清,检测血凝抑制抗体效价和中和抗体效价。结果 1针、2针组各3批无佐剂疫苗免疫小鼠的血凝抑制抗体效价和中和抗体效价分别小于1∶40和1∶200;2针、3针组各3批铝佐剂疫苗的血凝抑制抗体效价均大于1∶50,2针组20、30μg/ml血凝素铝佐剂疫苗的中和抗体效价均大于1∶200;2针、3针组20、30与45μg/ml血凝素铝佐剂疫苗的血凝抑制抗体效价和中和抗体效价差异均无统计学意义(P>0.05);且2针、3针组3批铝佐剂疫苗的血凝抑制抗体效价和中和抗体效价均明显高于相应的3批无佐剂疫苗。结论铝佐剂疫苗对小鼠的免疫原性明显优于无佐剂疫苗,且20、30μg/ml血凝素铝佐剂疫苗接种2针可获得理想的免疫效果。

Immunogenicity of Human Avian Influenza Split Vaccine to Mice |

Abstract: | Objective To observe the immunogenicity of human avian influenza split vaccine to mice.Methods The aluminum adjuvant-containing and adjuvant-free human avian influenza split vaccines with various hemagglutinin contents(20,30 and 45 μg / ml),three batches for each,were used for immunization of mice.Mice were divided various groups,10 for each,and were injected i.p.with 1,2 and 3 doses of the two kinds of vaccines respectively,0.5 ml for each,using those unimmunized as blank control.The interval between doses was one week.Serum samples were collected 21 d after the first dose and determined for HI and neutralizing antibody titers.Results The HI and neutralizing antibody titers of mice immunized with 1 and 2 doses of adjuvant-free vaccines were less than 1 ∶ 40 and less than 1 ∶ 200 respectively.However,the HI antibody titers of mice immunized with 2 and 3 doses of aluminum adjuvant-containing vaccine were more than 1 ∶ 50,and nuetralizing antibody titers with 2 doses of aluminum adjuvant-containing vaccine with hemagglutinin contents of 20 and 30 μg / ml were more than 1 ∶ 200.Neither HI nor neutralizing antibody titers showed significant difference in the mice immunized with 2 and 3 doses of aluminum-containing vaccines with hemagglutinin contents of 20,30 and 45 μg / ml(P > 0.05).However,both HI and neutralizing antibody titers of mice immunized with 2 and 3 doses of aluminum adjuvant-containing vaccine of three batches were significantly higher than those with adjuvant-free vaccine of the corresponding batches.Conclusion The immunogenicity of aluminum-containing vaccine was significantly superior to that of adjuvant-free vaccine,and the immune effects of 2 and 3 doses of aluminum-containing vaccine with hemagglutinin contents of 20 and 30 μg / ml were satisfactory. |
