摘    要:从提取溶剂及提取条件等方面对大豆抗原蛋白提取工艺进行了优化,采用单因素试验对提取溶剂及提取条件进行了分析研究,结果表明Tris-HCl缓冲液是理想的提取溶剂,提取温度、提取时间和料液比对大豆抗原蛋白提取产生一定的影响,其中提取温度的影响最为显著。通过Box-Benhnken中心组合设计和响应面分析法,确定了大豆抗原蛋白提取的理想工艺条件,即Tris-HCl缓冲液为溶剂,料液比1∶21.2(g∶mL),提取温度19.15℃,提取时间1.085h,在此条件下最大吸光度值为1.220,验证值为1.198,11S球蛋白提取得率为24.45%,较优化前提高了76.15%。

Abstract:Extraction of soybean antigen protein form soybean was optimized from extracting solvents and other extracting conditions. The results indicated that the Tris-HCl buffer was the best solvent. The results showed that the factors such as extracting temperature, extracting time, material and solvent ratio had effects on the extracting rate. Extracting temperature was the most significant factor for extracting rate compares with the other factors. The optimal conditions are described as follows: Tris-HCl buffer as extracting solvent, the ratio of material and solvent 1 ∶21.2,extracting temperature 19.15 ℃, extracting time 1.085 h by Box-Benhnken central composite design and Response Surface Methodology. Under the optimized condition, the highest extracting amount of absorbance form soybean antigen protein was 1.220,while the actual absorbance is 1.198. The total of 11S globulin extracting rate is 24.45 % which is 76.15 % higher than optimization of the former.
