摘    要:以泥鳅肉水解产物对血管紧张素转化酶(ACE)的抑制率、水解度(DH)以及水解产物的肽含量为指标,从碱性蛋白酶、碱性内切酶(Alcalase)、菠萝蛋白酶、胰蛋白酶、木瓜蛋白酶5种酶中筛选出碱性内切酶作为酶解泥鳅蛋白制备降血压肽的最适水解酶。在单因素试验基础上,采用响应面实验对该酶的酶解条件进行优化,得出最佳水解条件为:时间60min,pH 8.1,温度45℃,酶底物比(E/S)1 900U/g,固液比1∶15。在该条件下水解,得到酶解物的ACE半抑制质量浓度IC50值为0.22mg/mL。 更多还原

Abstract:Three indices(the degree of the angiotensin Ⅰ-converting enzyme(ACE) inhibitory activity,the degree of hydrolysis(DH) and the yield of peptide) of loach meat hydrolysates hydrolyzed by five different hydrolytic enzymes(alkali protease,alcalase,Bromelain,Trypsin and nematolyt) were evaluated.Alcalase was chosen as the optimal hydrolysis enzyme.On the basis of single factor experiment,response surface was used to optimizing the hydrolysate conditions.The result showed that the optimal enzymatic hydrolysis condition that could produce the protein hydrolysates with the highest activity on angiotensin-converting enzyme inhibitory activity hydrolyzed by alcalase was as follows: time 60 min,pH 8.1,temperature 45 ℃,(E/S)1 900 U/g,and substrate concentration 1∶15.Under such a condition,the IC50 of the ACE inhibitory peptides were 0.22 mg/mL.
