索骨丹总蒽醌和鞣质对细胞因子的影响

摘    要:观察索骨丹总蒽醌和鞣质对3种致炎细胞因子(IL-1β、IL-6、TNF-α)的影响,进而探讨其治疗炎症性疾病的部分机制。将体外培养的人单核细胞分为脂多糖组、不同浓度的含药组及阴性对照组。除对照组外,各组均加入相同量的脂多糖后继续培养12h,收集上清液,采用ELISA法通过酶标仪测得各组溶液的吸光度值,再计算出各组细胞因子的浓度。结果发现:索骨丹总蒽醌除0.50mg·mL-1组外,0.75~1.50mg·mL-1各组均能降低IL-1β含量(P0.05);索骨丹总蒽醌在1.00~1.50mg·mL-1范围时可降低IL-6含量(P0.05);索骨丹总蒽醌各浓度对TNF-α均有显著性抑制作用(P0.05)。索骨丹鞣质各浓度对TNF-α均有显著性抑制作用(P0.05)。表明,索骨丹提取物可通过减少TNF-α、IL-1β和IL-6的产生和释放而发挥治疗作用。

Abstract:The influence of anthraquinones and tannins from Rodgersia aesculifolia Batal.on cytokines was observed to explore the mechanism of anti-inflammatory.Human monocytes in vitro culture were divided into the lipopolysaccharide(LPS) group,the different concentrations of medicated groups and the negative control group.Each group was added the same amount of LPS.The supernatant after 12 h was collected and the absorbance value was measured for each solution by ELISA method,and then the cytokine concentration was calculated.It was found that anthraquinones of Rodgersia aesculifolia Batal.groups from 0.75 mg · mL-1 to 1.50 mg· mL-1 could reduce the content of IL-1β(P<0.05),from 1.00 mg · mL-1 to 1.50 mg · mL-1 could reduce the content of IL-6(P<0.05),from 0.50 mg · mL-1 to 1.50 mg· mL-1 could reduce the content of TNF-α(P<0.05).Every concentration of tannins from Rodgersia aesculifolia Batal.could significantly inhibit TNF-a(P <0.05).The results showed that extracts of Rodgersia aesculifolia Batal.could reduce the producing and releasing of TNF-a,IL-1fβ and IL-6 and thus played a role of treatment.
