摘    要:目的:通过人体新鲜粪便酵解模型研究广叶绣球菌多糖在体外的酵解特性,以便为其体内的消化特性研究提供理论支持。方法:制备广叶绣球菌多糖和人体粪便菌群混合液,在无氧环境下发酵0、1、2、6、12、24、48 h后,检测酵解液中的OD600值、pH值,总糖、还原糖、短链脂肪酸(乙酸、丙酸、丁酸)含量,乳酸杆菌、双歧杆菌和大肠杆菌数量。结果:在发酵过程中,酵解液的总糖含量显著下降(P<0.05),双歧杆菌数量显著上升(P<0.05);OD600值、还原糖含量、短链脂肪酸含量以及乳酸杆菌和大肠杆菌数量都在不同的时间点出现显著升高(P<0.05),pH值则出现显著下降(P<0.05),而随着时间延长,这些指标又呈现相反的变化。结论:广叶绣球菌多糖不仅能够被粪便菌群酵解利用,而且可以促进乳酸杆菌和双歧杆菌的生长,产生大量乙酸、丙酸和丁酸,并抑制大肠杆菌的生长。

Abstract:Objective:In order to provide theoretical support for the study of digestion characteristics in vivo,the in vitro fermentation characteristics of Spiraea latifolia polysaccharides were studied by using human fresh fecal fermentation model.Methods:The mixture of Spiraea latifolia polysaccharides and human fecal flora was prepared.After fermentation in anaerobic environment for 0,1,2,6,12,24 and 48 h,the OD600 value,pH value,total sugar,reducing sugar,short chain fatty acid(acetic acid,propionic acid,butyric acid)content,Lactobacillus,Bifidobacterium and Escherichia coli were detected.Results:During the fermentation process,the total sugar content decreased significantly(P<0.05),and the number of Bifidobacterium increased significantly(P<0.05);the OD600 value,reducing sugar content,short chain fatty acid content,Lactobacillus and Escherichia coli number increased significantly at different time points(P<0.05),while the pH value decreased significantly(P<0.05),and with the extension of time,these indicators showed opposite changes.Conclusion:The polysaccharide of Spiraea latifolia can not only be used by fecal microflora,but also can promote the growth of Lactobacillus and Bifidobacterium,produce a large amount of acetic acid,propionic acid and butyric acid,and inhibit the growth of Escherichia coli.
