摘 要: | 研究了重组海参(Stichopus japonicus)溶菌酶(SjLys)可溶性表达的发酵条件。采用多因素正交优选法,以摇瓶培养方式从培养基组分、发酵条件两个方面对重组海参溶菌酶进行研究,构建了高效分泌型表达重组海参溶菌酶工程菌。研究结果发现,30℃、140r/min、IPTG浓度0.3mmol/L、诱导后继续培养14h,目的蛋白分泌表达量可达到30~40mg/L,且产物以大量可溶性蛋白存在。

Expression and characterization of recombinant lysozyme from sea cucumber Stichopus japonicus |

Abstract: | Fermentation and characterization of the genetically engineered bacteria E.coli Rosetta(DE3)pLysS/pET-32a(+)-SjLys was studied to express the recombinant lysozyme from the sea cucumber Stichopus japonicus(rSjLys).The rSjLys was expressed in soluble form via optimizing fermentation media and conditions.The optimal medium composition for the engineering strain was determined by orthogonal test and the product of the rSjLys was purified by Ni 2+metal affinity chromatography.The production yield of the rSjLys(almost soluble protein) was obtained 30-40mg/L at the induction temperature 30 ℃,rotational speed 140r/min,induction concentration of IPTG 0.3mmol/L and culture time 14hafter induction. |
