摘    要:利用HPLC法对红曲霉发酵样品中MonacolinK的测定方法进行了研究。以V (甲醇 )∶V(0 18%磷酸 ) =77∶2 3为流动相 ,流速 0 6mL/min ,检测波长 2 3 7nm ,用外标法进行定量 ,相对标准偏差和平均回收率分别为 0 .2 7%和 98 92 %。用甲醇提取红曲米样品 ,摇床振荡 (3 0℃ ,2 0 0r/min) 2 5~ 3h可将MonacolinK提取完全。红曲霉摇瓶发酵产生的MonacolinK有 5 0 %以上存在于发酵滤液中 ,因此可以直接取滤液进行分析。该方法简便有效 ,适用于功能性红曲系列产品的科研开发及质量控制

Abstract:The method which determines the content of Monacolin K in fermentation samples of Monascus sp. by HPLC was studied Methanol:0 18% phasphate=77∶23(v/v) is used as mobile phase,flow rate 0 6 mL/min,wavelength of detection 237 nm The content of Monacolin K is determined by external standard method The relative standard deviation and average recovery are 0.27%?98.92% respectively. Red koji rice is extracted by methanol, and Monacolin K could be completely extracted by shaking for 2.5~3h with the revolving rate of 200 r/min and the temperature at 30℃. It is found that there are more than 50% Monacolin K existing in the culture filtrate of Monascus sp. Thus the filtrate could be directly used to determine Monacolin K. The experimental results show that the method is simple, effective, and it could be used for the research work and quality control of functional products derived from Monascus sp.
