摘    要:建立了以羧甲基-β-环糊精(CM-β-CD)作为手性流动相添加剂,高效液相色谱拆分标题化合物的方法。系统考察了流动相中添加剂的种类及浓度、有机修饰剂、pH、三乙胺的含量和流速等因素对拆分的影响。色谱分离条件:流动相(V(甲醇)∶V(乙腈)∶V(水)=15∶70∶15),CM-β-CD浓度为0.6 g/L,三乙胺含量为0.14%(V/V),pH为3.92,流速为0.2mL/min,检测波长214 nm,药物对映体得到良好分离。

Abstract:Separation of etilefrine hydrochloride enantiomers by HPLC was established using the carboxymethyl-β-cyclodextrin(CM-β-CD in short) as chiral mobile phase additive.The influences of type and concentration of chiral mobile phase additives,the organic modifier,the pH value,the concentration of triethylamine and the flow rate on the enantiomers separation were investigated.The beest separation was achieved when the mobile phase consist of(V(methanol)∶V(acetonitrile)∶V(water)=15∶70∶15),containing 0.6 g·L-1 CM-β-CD and 0.14%(V/V) triethylamine,pH 3.92,at the flow rate of 0.2 mL·min-1.The detection wavelength was set at 214 nm.
