柿子皮类胡萝卜素的超声提取及抗氧化活性的研究

摘    要:以柿子皮为原料,以类胡萝卜素的含量为试验指标,研究超声波提取柿子皮中类胡萝卜素的工艺,并对提取物进行了分离纯化和抗氧化性能的初步探讨。通过单因素和正交试验得出以丙酮为提取溶剂,在柿子皮粉碎度为40目、超声频率40kHz、提取两次条件下,柿子皮中类胡萝卜素的最佳提取工艺为:料液比1∶25(g∶mL)、超声功率400W、超声时间20min。在此条件下柿子皮提取物中类胡萝卜素浓度为0.3872μg/mL,分离纯化物β-胡萝卜素的得率为3.99mg/kg(柿子皮),叶黄素为19.84mg/kg(柿子皮),表明柿子皮具有较高的研究和开发价值。抗氧化试验结果表明:柿叶提取液对超氧阴离子(O2-.)和羟自由基(.OH)均具有较强的抑制作用,并且随着提取液浓度的增加而有增强的趋势。表明柿子皮类胡萝卜素是一种很有潜力的天然、安全、高效的抗氧化剂。

Abstract:The technology of extracting Carotenoid by ultrasonic wave from Persimmon husk,the separate purification and their antioxidant activities were studied simultaneously,taking the content of Carotenoid as a criterion.The best technical conditions for extraction Carotenoid obtained through the single factor and orthogonal test was as follows:the ratio of material and solution as 1∶25,ultrasonic power was 400W,extracting time was 20min,Persimmon husk granularity was 40,ultrasonic wave frequency was 40kHz,two extraction times,and the extraction solvent was acetone.Under this condition the concentration of Carotenoid was 0.387 2μg/mL and the extraction rate of β-Carotene was 3.99mg/kg and lutein was 19.84mg/kg,indicating it would be valued for research and exploitation.The results showed that Carotenoid could inhibit the reaction of O2-·and ·OH.With the increase of the extracting concentration,the scavenging abilities also increased.Carotenoid from the Persimmon husk was a potential product of natural,safe and effective antioxidants.
