浓香型白酒窖泥中总DNA提取方法的研究

摘    要:本研究针对5种白酒窖泥总DNA提取方法(预处理+DNA提取液+SDS法,改进的化学裂解法,CTAB+ SDS+反复冻融,SDS细胞裂解法,玻璃珠+CTAB+溶菌酶法),探索适合微生物多样性研究的细菌16S rDNA和真菌ITS-5.8S rDNA的PCR扩增的窖泥总DNA提取方法.结果表明,采用玻璃珠+CTAB+溶菌酶法和化学裂解法提取DNA无明显降解、重复性好,可用于后续细菌16S rDNA和真菌ITS-5.8S rDNA的PCR扩增.

Abstract:In order to extract DNA from Gujing-flavor liquor pit mud under simple and easy experimental terms for follow-up 16S rDNA and ITS-5.8S rDNA PCR analysis, five methods(pretreatment+extraetion buffer+ SDS method, grinding + freezing + thawing method, CTAB + SDS + freeze-- thaw method, SDS method and beads+lysozyme+ CTAB method) were employed and compared,it can be concluded that the beads+lysozyme+SDS method and grinding+ freezing+ thawing method are expected to be adopted as rapid and reliable protocols for further microbial molecular study of Pit Mud.
