高效氯氰菊酯降解菌的分离与鉴定及其降解条件优化

摘    要:从长期施用高效氯氰菊酯农药的菜园土壤中,分离到一株能降解高效氯氰菊酯的细菌GCN019.经生理生化和16S rDNA分析,将菌株GCN019鉴定为荧光假单胞菌(Pseudomonas fluorescens).应用Plackett-Burman试验设计确定了影响该菌株降解高效氯氰菊酯的主要因素,利用响应面分析法优化了其降解条件.结果表明,降解体系中高效氯氰菊酯浓度和接种量及初始pH是影响其降解的主要因素;优化条件下高效氯氰菊酯的降解率可达到73.94%,与所建立的模型预测值(74.30%)相吻合.

Abstract:Strain GCN019 with high ability to degrade beta-cypermethrin was isolated from garden soil which contaminated by beta-cypermethrin for a long term. The strain was identified as Pseudomonas fluorescens based on the results from its physiological-biochemical characteristics and phylogenetic similarity of 16S rDNA gene sequences. Then, the main factors influencing beta-cypermethrin degradation were investigated by using Plackett-Burman design, and the degradation condition was optimized with response surface methodology. It was found that beta-cypermethrin concentration, inoculum volume and initial pH value were the major factors influencing beta-cypermethrin degradation. In addition, the degradation rate of beta-cypermethrin under the optimized condition reached 73.94%, which was similar to the predictive value from response surface model (74.30%).
