摘    要:将Plackett-Burman设计法和响应面分析法相结合,对假单胞菌M18合成吩嗪-1-羧酸的发酵条件进行了优化,结果表明,初始葡萄糖浓度和种龄是影响吩嗪-1-羧酸发酵效价的主要因素。优化后的培养条件为葡萄糖0.87%,甘油0.6%,乙醇0.8%,黄豆粉4%,蛋白胨0.4%,pH7.0,瓶装量40/250,接种量4%,种龄10.3h,温度28℃。在此条件下,吩嗪-1-羧酸的发酵效价为1432mg/L,比优化前提高31.1%。利用优化条件在10L全自动发酵罐中进行了试验,吩嗪-1-羧酸的发酵效价为1418mg/L,平均合成速率达到23.63mg/(L·h),提高55.8%。

Abstract:Plackett-burman and response surface analysis methods were applied to optimize conditions of phenazine-1-carboxylic acid fermentation. The results indicated that phenazine-1-carboxylic acid yield was mainly influenced by concentration of initial glucose and age of inoculum. The obtained optimal fermentation conditions were glucose 0.87%, glycerol 0.6%, ethanol 0.8%, soybean meal 4%, peptone 0.4%, pH 7.0, liquid loading 40/250, inoculum 4%, age of inoculum. 10.3 h, temperature 28℃. Under these conditions, the phenazine-1-carboxylic acid yield was 1 432 mg/L, advanced 31.1%. The fermenter cultivation showed that the average synthesis rate of phenazine-1-carboxylic acid reached 23.63 mg/(L·h), advanced 55.8%.
