多元醇对绿色木霉纤维素酶热稳定性的影响

摘    要:
研究绿色木霉属纤维素酶的最适温度及最适pH值,探讨其在不同温度下的热失活情况,以及多元醇(海藻糖、聚乙二醇400、乙二醇、甘油)对其热稳定性的影响。结果表明,绿色木霉属纤维素酶最适温度为50~55℃,最适pH值为4.6;其在70℃时迅速失活,失活半衰期为3.2 min。多元醇的加入能在一定程度上改善其热稳定性,其中添加聚乙二醇400和海藻糖的混合物,能将其半衰期延长至5.9 min。

Abstract:
The optimum applicable temperature and pH value of cellulase (originated from trichoderma Viride) are studied. The thermal inactivition and the effect of polyols(trehalose, PEG400, glycol and glycerol)on thermal stability of cellulase are discussed. The results show that the optimum temperature and pH value of cellulase is 50~55 °C and 4.6 respectively. The cellulase inactivates quickly at 70 °C, and the half life time of inactivition is 3.2 min. The thermal stability of cellulase improves with the addition of polyols, and its half life time prolongs to 5.9 min at the presence of PEG 400 and trehalose mixture.
