柱层析法分离纯化大豆磷脂酰胆碱

摘    要:利用硅胶柱层析法,通过梯度洗脱对大豆粉末磷脂中的磷脂酰胆碱进行分离纯化.比较氯仿/甲醇和异丙醇/甲醇两个洗脱体系所得磷脂酰胆碱的纯度和回收率.梯度洗脱时,在氯仿/甲醇体系中氯仿:甲醇的比例为3:2(体积分数),异丙醇/甲醇体系异丙醇:甲醇的比例为1:1(体积分数)洗脱,是获得磷脂酰胆碱的最佳洗脱条件.结果表明:在氯仿,甲醇洗脱体系获得磷脂酰胆碱的纯度为97.0%、回收率为86.5%.异丙醇/甲醇洗脱体系获得磷脂酰胆碱的纯度为95.0%、回收率为83.0%.

Abstract:Separation and purification the phosphatidylcholine(PC)from soy lecithin powder by gradient elution using the silica gel column chromatography was stduied.Purity and recovery of phosphatidylcholine,which obtained from chloroform/methanol and isopropyl alcohol/methanol elution system were compared.The optimal conditions for elution of phosphatidylcholine were chloroform/methanol (3:2)and isopropyl alcohol/methanol (1:1)for chloroform/methanol and isopropyl alcohol/methanol system,respectively.The purities of...
