摘    要:测定了一种以自制的新型载体制备的固定化青霉素酰化酶的基本性质和各项动力学参数.固定化酶的稳定性与中科院微生物研究所用同种游离酶制备的纤维状固定化酶相当;其活性对温度和pH值的变化不如游离酶敏感;米氏常数、底物抑制常数、苯乙酸和6-氨基青霉烷酸的抑制常数分别为:0.679,749,2.24和92mmol/L.

Abstract:The properties of WTBX] Bacillus megarium penicillin amidase which is immobilized on a novel microbead carrier invented in the authors'laboratory were studied. The kinetic parameters were determined. The stability of the immobilized enzyme is the same as that of fibrous IME made from same free enzyme. The activity of the IME is not as susceptible to the changes of temperature or pH as that of free enzyme. The Michaelis Menton constant and the inhibition constants of the substrate, phenylacetic acid (PAA) and6-APA are 0.679, 749, 2.24 and 92 mmol/L, respectively.
