鼠李糖乳杆菌LP216高密度发酵培养基优化

摘    要:为提高鼠李糖乳酸杆菌LP216生产效益,通过单因素试验、Plackett-Burman(PB)试验和Box-Behnken(BB)试验,对菌株LP216发酵培养基进行优化。结果表明,最佳培养基配方为酵母提取物16 g/L、蛋白胨13 g/L、葡萄糖30 g/L、牛肉膏5 g/L、CH3COONa 4 g/L、吐温80 1.5 g/L、MgSO4·7H2O 0.25 g/L、柠檬酸二胺2.0 g/L、K2HPO4 2.0 g/L、MnSO4 0.2 g/L。在此优化条件下,菌株LP216活菌数达8.9×109 CFU/mL,是对照组(MRS培养基)活菌数(3.28×109 CFU/mL)的2.71倍。

Abstract:In order to improve the production efficiency of Lactobacillus rhamnosus LP216, the fermentation medium of strain LP216 was optimized by single factor tests, Plackett Burman (PB) tests and Box Behnken (BB) tests. The results showed that the optimum formula of medium was yeast extract 16 g/L, peptone 13 g/L, glucose 30 g/L, beef extract 5 g/L, CH3COONa 4 g/L, Tween 80 1.5 g/L, MgSO4·7H2O 0.25 g/L, citric diamine 2.0 g/L, K2HPO4 2.0 g/L and MnSO4 0.2 g/L. Under the optimal conditions, the viable counts of strain LP216 reached 8.9×109 CFU/ml, which was 2.71 times of control group (MRS medium) (3.28×109 CFU/ml).
