褐环乳牛肝菌液体发酵培养基的优化

摘    要:筛选褐环乳牛肝菌(Suillus luteus)最适培养基,以为后续的菌根及生态研究提供研究基础。通过单因素试验、Plackett-Burman试验、最陡爬坡试验及基于中心组合法的响应面试验,优化了褐环乳牛肝菌液体发酵培养基配方。结果表明,褐环乳牛肝菌最适碳源为果糖,最适氮源为牛肉膏,最佳褐环乳牛肝菌液体发酵培养基组成为牛肉膏7.9 g/L,β-环糊精16.8 g/L,抗坏血酸0.005 5 g/L,果糖20 g/L,CaCl2 0.1 g/L,NaCl 0.075 g/L,MgSO4 0.9 g/L,KH2PO4 1.0 g/L。采用最优培养基培养20 d可获得菌丝干质量7 656 mg/L。

Abstract:The optimal medium for Suillus luteus Gray was screened to provide a basis for the subsequent mycorrhizal and ecological studies. The liquid fermentation medium formula for S. luteus was optimized by single factor tests, Plackett-Burman tests, steepest climbing tests and response surface tests based on central composite methodology. The results showed that the optimal carbon source and nitrogen source were fructose and beef extract, respectively. The optimal liquid fermentation medium composition for S. luteus Gray were as follows: beef extract 7.9 g/L, β-cyclodextrin 16.8 g/L, ascorbic acid 0.005 5 g/L, fructose 20 g/L, CaCl2 0.1 g/L, NaCl 0.075 g/L, MgSO4 0.9 g/L, and KH2PO4 1.0 g/L. The dry mass of mycelial was 7 656 mg/L after culture for 20 d with the optimal medium.
