国产及进口龙血竭原料中总黄酮、龙血素A、龙血素B含量的测定

摘    要:比较国产及进口龙血竭原料中总黄酮、龙血素A、龙血素B的含量.龙血素A、龙血素B采用高效液相色谱法(HPLC)法测定,色谱柱安捷伦ZORBAX Eclipse XDB C18(4.6 mm×250 mm,5μm),流动相为乙腈-1%冰醋酸(40:60),流速1.0 mL/min,柱温25 ℃,检测波长为280 nm.总黄酮用50%乙醇超声提取,紫外-可见分光光度法测定.结果 龙血素A在0.0216~0.1948 μg范围内呈线性关系,回归方程为Y=-0.7836X+1.8251,r=0.9998,龙血素B在0.0201~0.1811μg范围内呈线性关系,回归方程为Y=-0.4785X +1.0548,r=0.9997,不同批次的龙血竭原料中龙血素 A 含量在0.021%~0.177%,龙血素 B 含量在0.018%~0.075%,总黄酮含量在4.287%~8.531%.结论 该方法快速、准确,为龙血竭及其原料的质量控制提供了科学依据.

Determination of Total Flavone,Loureirin A and Loureirin B in Domestic and Imported Dracaena Cochinchinensis Raw Materials

Abstract:In order to compare total flavone,loureirin A and loureirin B contents in domestic and imported dracaena cochinchinensis raw materials, loureirin A and loureirin B contents were determined by HPLC with Agilent ZORBAX Eclipse XDB C18(4.6 mm×250 mm,5μm)under 280nm UV detection wavelength. The total flavone was extracted by 50% ethanol using ultrasonic and determined by UV-Vis spectrophotometry. The results showed that the loureirin A content showed good linearity in the range of 0.0216~0.1948 μg,regression equation: Y=-0.7836X +1.825 1,correlation coefficient(r)=0.999 8. The loureirin B content showed good linearity in the range of 0.0201~0.1811 μg,regression equation: Y=-0.4785X +1.0548,correlation coefficient(r)=0.999 7.The content of loureirin A ranged from 0.021%~0.177% and the content of loureirin B ranged from 0.018%~0.075%. The content of total flavone ranged from 4.287%~8.531%. The determination method of total flavone,loureirin A and loureirin B is simple and repeatable, and can be used for the quality control of dracaena cochinchinensis raw materials.
