酶解对乌骨鸡多肽抗氧化活性影响的研究

摘    要:研究了不同的酶及酶解条件对乌骨鸡多肽抗氧化能力的影响,以得到最优的酶解条件及较高抗氧化能力的多肽。结果表明,在温度为50℃、酶用量[E/S]8000U/g、底物浓度为7%时,胰蛋白酶水解乌骨鸡多肽可以获得更高抗氧化能力的多肽;进一步对胰蛋白酶水解条件正交实验优化,结果表明,胰蛋白酶水解的最优条件为温度50℃、pH8.5、酶用量[E/S]6000U/g、反应时间6h及底物浓度9%,此时有最高的抗氧化能力,在33.3μg/mL乌骨鸡多肽浓度时能清除45.41%的羟自由基,比同一浓度条件下的VC的羟自由基清除率高了4%。

Abstract:In this paper,effects of different enzymes and hydrolysis conditions on the antioxidant activity of black-bone silky fowl peptides were studied to acquire the best hydrolysis conditions and peptides with higher antioxidant activity. The result showed that under the temperature of 50℃, E/S 8000U/g, pH 8.5, substrate concentration of 7% using trypsinase,peptides with higher antioxidant activity could be obtained.Further research on hydrolysis conditions of trypsinase using orthogonal design method showed that the best hydrolysis conditions was when the temperature was 50℃, pH 8.5, E/S 60000U/g, reaction time 6h and the substrate concentration 9%. Under that condition, hydrolysis products of black-bone silky fowl peptides had the most highly antioxidant activity of which 33.3μg/mL concentration could removal 45.41% hydroxyl radical,4% higher than V_C.
