中药锦灯笼中酸浆苦素类化合物含量测定

摘    要:以中药锦灯笼中抗炎活性成分4,7-二去氢新酸浆苦素B为对照品,用HPLC法对该药材进行了含量测定研究。Agilent ZORBAX Eclipse XDB C18色谱柱(4.6×250mm,5μm),以乙腈-0.1%磷酸水溶液(40:60)为流动相,流速1.0mL.min-1,检测波长353nm,柱温为室温。该物质在3.27×10-3~2.62×10-2μg呈良好的线性关系,平均回收率为99.1%。该方法简单易行,结果稳定、可靠,为该药材质量标准的建立提供依据。

Abstract:A high-pressure liquid chromatography(HPLC) method with ultraviolet detection was developed for the measure of 4,7-didehydroneophisalin B in Physalis alkekengi.Chromatographic separation was carried on a ZORBAX Eclipse XDB C18 column(4.6×250mm,5μm),with an isocratic mobile phase consisting of acetonitrile-0.1% phosphate(40:60,v/v),a flow rate of 1.0mL/min,with UV detection(353nm).A linear relationship was obtained between peak area and drugs concentration in the range studied(3.27×10^-3~2.62×10^-2μg for 4,7-didehydroneophisalin B).The equation for regression line were Y=1×10^-6X+489.11 for 4,7-didehydroneophisalin B,and correlation coefficients were〉0.999.This HPLC assay for 4,7-didehydroneophisalin B was sensitive and accurate,and provided a reliable determination of Physalis alkekengi,thus the quality of this plant was ensured.
